Apr\u00e8s les laboratoires sur puce d\u00e9velopp\u00e9s depuis plusieurs ann\u00e9es, les chercheurs et les entreprises sp\u00e9cialis\u00e9es dans les microtechniques travaillent d\u2019arrache-pied pour lever les verrous techniques et biologiques \u00e0 l\u2019industrialisation des organo\u00efdes sur puce.<\/p>\n<p>\u00ab <em>Initialement, des dispositifs microfluidiques, \u00e9galement appel\u00e9s laboratoires sur puce, \u00e9taient d\u00e9velopp\u00e9s par des technologues pour faire de la biochimie avec de tr\u00e8s petits volumes de r\u00e9actifs. Vers le milieu des ann\u00e9es 2000, ils ont implant\u00e9 des couches de cellules sur leurs puces pour mimer certaines fonctions physiologiques <\/em> \u00bb, pr\u00e9cise Xavier Gidrol, chef de service au CEA-Irig (1). \u00ab <em>De leur c\u00f4t\u00e9, les chercheurs sp\u00e9cialistes des cellules souches ont cr\u00e9\u00e9 des objets biologiques assez complexes, les organo\u00efdes, pour comprendre le d\u00e9veloppement des organes ou des organismes <\/em> \u00bb. Ces derniers sont con\u00e7us en trois dimensions, cultiv\u00e9s en suspension dans un hydrogel et obtenus \u00e0 partir de cellules souches. Ils comportent donc diff\u00e9rents types de cellules pr\u00e9sents dans un organe et ex\u00e9cutent au moins une partie de ses fonctions.<\/p>\n<p>En somme, selon Xavier Gidrol \u00ab <em>si on souhaite \u00e9tudier un mod\u00e8le biologique qui ressemble vraiment \u00e0 un organe, il faut cr\u00e9er un organo\u00efde. Mais si on veut contr\u00f4ler de mani\u00e8re reproductible sa forme et sa taille, monitorer ses fonctions correctement, etc., il faut le mettre sur puce. Il s\u2019agit donc de r\u00e9unir le meilleur des deux mondes <\/em> \u00bb. Ces diff\u00e9rents dispositifs servent aussi bien la recherche fondamentale, l\u2019industrie pharmaceutique, que la m\u00e9decine personnalis\u00e9e, voire la production de tissus pour la m\u00e9decine r\u00e9g\u00e9n\u00e9ratrice. Le march\u00e9 annonc\u00e9 est immense\u2026 Cependant, il faut r\u00e9pondre \u00e0 des exigences telles que la reproductibilit\u00e9, la fiabilit\u00e9, la fabrication en s\u00e9rie \u00e0 co\u00fbt \u00e9conomiquement viable, la facilit\u00e9 de transport et d\u2019utilisation.<\/p>\n<p>Autant de d\u00e9fis qui poussent le CEA \u00e0 rechercher constamment de nouveaux mat\u00e9riaux et nouveaux process d\u2019assemblage concernant trois axes principaux : les dispositifs technologiques, les syst\u00e8mes biologiques issus de cellules souches et les capteurs &#8211; miniaturis\u00e9s et embarqu\u00e9s sur les puces. \u00ab <em>Nous anticipons cela d\u00e8s le d\u00e9but par le choix des mat\u00e9riaux et, c\u2019est tr\u00e8s important, des proc\u00e9d\u00e9s d\u2019assemblage. Au stade du laboratoire, nous travaillons avec diff\u00e9rentes techniques pour faire du prototypage rapide : micro-usinage ou gaufrage \u00e0 chaud. Mais d\u00e8s que le design de la puce est fig\u00e9, nous passons \u00e0 la plasturgie industrielle. Nous r\u00e9alisons un moule et injectons les mat\u00e9riaux, de mani\u00e8re \u00e0 pouvoir faire des milliers de fois le m\u00eame composant, au besoin <\/em> \u00bb, explique Fabrice Navarro.<\/p>\n<p>\u00ab <em>Seuls quelques organes sur puce sont actuellement commercialis\u00e9s.
